What is a Generic Drug?

Description

Talzenna (Talazoparib) is used as an anti-cancer treatment for certain breast and prostate cancers. More specifically, it targets cancer types with BRCA1 or BRCA2 gene mutations. As a PARP inhibitor, Talzenna blocks the PARP enzyme, thereby preventing BRCA-mutated cancer cells from repairing their DNA and causing them to die.

Directions

Depending on factors such as your kidney function and your body’s response to the medication, your healthcare provider may prescribe Talzenna in one of the following strengths: 0.1 mg, 0.25 mg, 0.35 mg, 0.5 mg, 0.75 mg, or 1 mg.

This oral capsule should be taken exactly as prescribed by your doctor. Swallow whole with or without food and do not open or dissolve. If you vomit or miss a dose of Talzenna, wait until your next scheduled dose.

Your prescribed Talzenna dosage also varies depending on the condition being treated. The dosing schedules mentioned below are typical but may vary by individual:

Breast Cancer

Take a 1 mg dose of Talzenna once daily as long as the treatment remains effective. You may need to discontinue use if side effects make it intolerable or unsafe.

Prostate Cancer

Take a 0.5 mg dose once daily as long as the treatment remains effective. You may need to discontinue use if side effects make it intolerable or unsafe. Talzenna and enzalutamide are typically combined for the treatment of prostate cancer, and you may need additional medical or surgical treatments.

Ingredients

The active ingredient in Talzenna is talazoparib tosylate.

Inactive ingredients are silicified microcrystalline cellulose, hypromellose, yellow iron oxide, red iron oxide, titanium dioxide, shellac, black iron oxide, potassium hydroxide, ammonium hydroxide, and propylene glycol.

Contraindications

According to the FDA, there are no contraindications. However, your doctor will determine whether you can take Talzenna on an individual basis.

Cautions

  • Before taking Talzenna, inform your doctor of your complete medical history, including:
    • Other medical conditions, especially kidney issues, blood disorders, prior cancer treatments, allergies, pregnancy, breastfeeding status, and fertility concerns.
    • Your allergies. Do not take Talzenna if you’re allergic to one of its ingredients. Seek immediate medical care if you experience allergy symptoms such as rash, itching, swelling, or trouble breathing.
    • Your pregnancy and breastfeeding status. Avoid pregnancy during treatment as Talzenna can harm an unborn baby. Do not breastfeed while taking Talzenna or for at least 1 month after your last dose.
    • A list of medications you’re taking, including heart or blood pressure drugs, antibiotics, antifungals, chemotherapy, medications that affect blood count, and herbal or vitamin supplements. Some medications can increase Talzenna levels or lower blood counts further, causing serious side effects.
  • Reduced kidney function may impact the levels of Talzenna in your body. In this case, your doctor may prescribe a lower Talzenna dosage.
  • When you take Talzenna, regular blood tests are required to monitor for low red, white, or platelet counts, as well as rare cases of myelodysplastic syndrome (MDS) or acute myeloid leukemia (AML). If any issues are detected, your doctor will adjust your dose or stop the treatment.

Side Effects

When taking Talzenna alone:

Fatigue, nausea, vomiting, headache, and loss of appetite are relatively common but often manageable. More serious side effects requiring medical attention include low red blood cell count (anemia), low white blood cell count (increased risk of infection), low platelet count (increased risk of bleeding and bruising), myelodysplastic syndrome (MDS), acute myeloid leukemia (AML) (rare), and fetal harm.

When taking Talzenna in combination with enzalutamide:

Tiredness, weakness, nausea, decreased appetite, dizziness, and changes in sense of taste are reported relatively frequently. More serious side effects requiring medical attention include low red blood cell count (anemia), low white blood cell count (increased risk of infection), low platelet count (increased risk of bleeding and bruising), bone injuries and fractures, changes in blood minerals, and fertility problems in males.
