Piqray (alpelisib) is a prescription medication for treating adults who have:

  • hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative, advanced or metastatic breast cancer with PIK3CA-mutated gene that has progressed on or after endocrine-based treatment

Piqray comes in 50 mg, 150 mg, and 200 mg tablets for oral administration.

Directions

Piqray should be taken as your healthcare provider has directed. Read the medication guide thoroughly that accompanies your prescription.

Piqray dosing:

Your healthcare provider will determine an appropriate dose for you. The recommended dose is 300 mg by mouth once daily. Take with food.

Store Piqray tablets at room temperature between 68°F and 77°F (20°C and 25°C) and away from children.

Any questions or concerns may be referred to your healthcare provider or pharmacist.

Ingredients

Alpelisib is the active ingredient in Piqray.

Hypromellose, magnesium stearate, mannitol, microcrystalline cellulose, sodium starch glycolate, iron oxide black, iron oxide red, macrogol/polyethylene glycol (PEG) 4000, talc, and titanium dioxide are the inactive ingredients.

Contraindications

Piqray should not be given to patients who are allergic to any of its ingredients.

Cautions

  • Before you start treatment with Piqray, inform your healthcare provider:
    • Of all the medications you take, including prescription and nonprescription medications, vitamins, herbals, and supplements
    • Of your allergies
    • Of your current health problems
    • Of your past medical history
    • If you are pregnant or breastfeeding
  • The following can occur during treatment with Piqray:
    • Hypersensitivity that may be severe
    • Cutaneous adverse reactions that may be severe
    • High blood sugar levels
    • Pneumonitis and interstitial lung disease
    • Diarrhea or colitis
    • Harm to a fetus. Females of childbearing potential and males with female partners who are of childbearing potential should be advised to use effective contraceptive methods during treatment with Piqray and for one week after receiving the final dose.
  • Breastfeeding is not advised.

Side Effects

Piqray side effects include but are not limited to high blood sugar levels; elevated creatinine; diarrhea; rash; decreased blood sugar levels, lymphocytes, and hemoglobin; increased gamma-glutamyl transferase (GGT), liver enzymes, and lipase; nausea; fatigue; decreased appetite; swelling/sores of the mouth; vomiting; weight loss; decreased calcium levels; hair loss; and prolonged blood clotting time. Contact your healthcare provider right away if you have bothersome or persistent side effects.
