What is a Generic Drug?

Description

Inclusig (ponatinib) is a medication prescribed to treat adults with Philadelphia Chromosome-Positive Acute Lymphoblastic Leukemia and Chronic Myeloid Leukemia.

Inclusig 10 mg, 15 mg, 30 mg, and 45 mg tablets are available for oral administration.

Directions

Inclusig dosage: Inclusig should be taken as your doctor has directed. Your doctor will tell you how much medication you should take.

Store Inclusig tablets at temperatures between 20°C to 25°C (68°F to 77°F); keep away from children.

Ingredients

Ponatinib is the active ingredient in Inclusig.

Lactose monohydrate, microcrystalline cellulose, sodium starch glycolate (type B), colloidal silicon dioxide, magnesium stearate and a tablet coating of talc, polyethylene glycol, polyvinyl alcohol, and titanium dioxide are the inactive ingredients included.

Contraindications

Inclusig does not have any contraindications per the most recent Prescribing Information.

Cautions

  • Inclusig is associated with the following FDA Boxed Warning:
    • Arterial occlusive events (AOEs), such as death, heart attack, stroke, stenosis of large arterial vessels in the brain, severe peripheral vascular disease, and necessary urgent revascularization procedures, have occurred in patients with and without cardiovascular risk factors, including those younger than 50, who were treated with Inclusig. Patients should be monitored for suspected AOEs. Inclusig therapy should be interrupted or discontinued depending on the severity of the AOE. The benefits and risks should be considered before restarting Inclusig.
    • Venous blood clots have occurred in patients taking Inclusig. Patients should be monitored, and interruption or discontinuation of Inclusig should occur depending on the severity of the blood clotting event.
    • Heart failure and death have occurred in patients taking Inclusig. When clinically indicated, monitoring and management of heart failure should occur. If heart failure develops or worsens, interruption or discontinuation of Inclusig should occur.
    • Liver toxicity, liver failure, and even death have occurred in patients receiving Inclusig. Monitoring of liver function tests should be performed, and interruption or discontinuation of Inclusig should occur based on the severity of the liver issue.
  • Discuss the following with your doctor before you begin taking Inclusig:
    • All the medications you take, including prescription and nonprescription medications, supplements, and vitamins
    • Any allergies you have
    • Any current health problems you have
    • Your past medical history
    • If you are pregnant or breastfeeding
  • Inclusig is associated with the following warnings and precautions:
    • High blood pressure
    • Pancreatitis
    • Nerve damage
    • Eye issues
    • Major bleeding
    • Fluid retention
    • Abnormal heart rhythms
    • Reduction in bone marrow activity
    • Tumor lysis syndrome
    • Reversible posterior leukoencephalopathy syndrome
    • Delayed wound healing and stomach/intestinal perforation
    • Fetal harm; females of reproductive potential should be advised to use effective contraception while being treated with Inclusig and for 3 weeks after receiving the last dose of treatment.

Side Effects

Inclusig side effects when taking the medication by itself include but are not limited to: rash and related conditions, abdominal pain, joint pain, headache, dry skin, constipation, high blood pressure, fatigue, fluid retention and swelling, nausea, fever, pancreatitis/lipase elevation, major bleeding, anemia, liver issues, AOEs, and abnormal labs (reduction in platelets, neutrophils, and white blood cells)

Side effects when taking Inclusig with chemotherapy include but are not limited to: liver issues, rash and related conditions, joint pain, headache, fever, abdominal pain, constipation, nausea, fatigue, oral mucositis, high blood pressure, pancreatitis/lipase elevation, nerve damage, major bleeding, febrile neutropenia, fluid retention and swelling, vomiting, tingling/numbness, abnormal heart rhythms, and abnormal labs (decreased white blood cells, neutrophils, platelets, lymphocytes, and hemoglobin and increased lipase and alanine aminotransferase)
