Orforglipron is classified as a GLP-1 receptor agonist, a type of medication that acts on the same receptors as a hormone your body naturally produces after eating. By activating these receptors, Foundayo helps curb appetite, promotes satiety, and slows digestion, supporting patients in reducing their overall calorie intake and sustaining weight loss over the long term.

Foundayo is a small-molecule oral tablet, which means it does not require fasting, special timing, or water restrictions before or after taking it. It can be taken once daily at any hour, with or without a meal.

Directions

Foundayo treatment begins at a starting dose of 0.8 mg once daily by mouth. To allow the body to adjust and minimize gastrointestinal discomfort, the dose is stepped up gradually, with increases no sooner than every 30 days, up to a maximum dose of 17.2 mg once daily, depending on how well the medication is tolerated and how the patient responds.

Tablets must be swallowed whole and should not be split, crushed, or chewed. There are no food or fluid requirements associated with taking Foundayo, it may be taken at any point during the day, regardless of meals.

If a dose is skipped, take it when remembered, provided you do not take two doses within the same calendar day. Should treatment be paused for a week or more, speak with your prescriber before resuming.

Ingredients

The active ingredient in Foundayo is orforglipron.

The tablet also contains the following inactive ingredients: copovidone, crospovidone, magnesium stearate, microcrystalline cellulose, and anhydrous sodium carbonate. The film coat is composed of polyethylene glycol, polyvinyl alcohol, talc, and titanium dioxide.

Cautions

Foundayo is contraindicated in patients who have a personal or family history of medullary thyroid carcinoma (MTC) or a diagnosis of Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).

This medication should not be taken by anyone with a known allergy or serious hypersensitivity reaction to orforglipron or to any component of the formulation.

Before starting treatment, advise your doctor of any history of pancreatitis, kidney disease, significant liver impairment, delayed gastric emptying (gastroparesis), or diabetic retinopathy.

Patients using oral contraceptives should be aware that Foundayo may decrease their effectiveness. An additional or alternative form of birth control may be advised during the first 30 days of treatment and for 30 days following each dose escalation.

Use during pregnancy is not advised, and breastfeeding while on this medication is not recommended. Always give your healthcare provider a full picture of your health history, current medications, and any known allergies before beginning Foundayo.

Side Effects

Commonly reported side effects with Foundayo use include nausea, stomach pain or cramping, acid reflux, constipation, loose stools, gas, bloating, vomiting, indigestion, burping, headache, tiredness, and temporary hair thinning.

More serious reactions that have been associated with this medication include pancreatitis, gallbladder complications, kidney injury, hypoglycemia when combined with other glucose-lowering agents, severe hypersensitivity reactions, and visual disturbances in patients managing type 2 diabetes.

If you develop severe or prolonged abdominal pain, any signs of a serious allergic reaction, or any other symptoms that feel unusual or persistent, discontinue use and seek medical attention promptly. The above does not represent a comprehensive list of every potential side effect.
